A Correlative Study of Gleason Grade with Proliferative Activitives (DNA content, Ag-NORs and PCNA) and Oncoprotein Expression (c-erbB, c-H-ras oncogenes) in Prostatic Adenocarinoma)

Abstract
In prostate adenocarcinoma, it has been well known that the prognosis of this tumor is related to the histologic differentiation. To evaluate the relationship between the histologic Gleason¡¯s grade and the proliferative activity as well as
oncoprotein expression, the author attempted flow cytometric DNA analysis, argyrophilic staining for Ag-NORs AND immunohistochemical staining for PCNA, and c-erbB-2, and c-H-ras oncoprotein on 40 cases of prostatic denocarcinoma with formalin fixed, paraffin-embedded tissue.

The results obtained were as follows;
1. Among 40 prostatic adenocarcinomas, 23 cases (57.5%) were aneuploidy in DNA pattern. The averages of S-phase fractions of diploid and aneuploid group were 3.77% and 9.66%, respectively. No correlation was observed between histologic
Gleason¡¯s
grade and DNA pattern.
2. The positivity of proliferating cell nuclear antigen (PCNA) was closely related to the Gleason¡¯s grade. Also a positive relation was observed between Gleason¡¯s grade and expressions of c-erbB-2 and c-H-ras oncoproteins, but it was
statistically
not
significant.
3. 3. There was no correlation between Ag-NORs or PCNA and S-phase fraction.
4. The number of Ag-NORs, the positive rate of PCNA, expression of c-erbB-2 and c-H-ras oncoprotein. and S-phase fraction were higher in aneuploid group than diploid group.
